Aurintricarboxylic Acid Suppresses Hepatitis B Virus Replication by Inhibition of RNase H Activity

چکیده

Background Chronic hepatitis B virus (HBV) infection is a global health threat for which there an urgent need to develop novel therapeutics. Methods Aurintricarboxylic acid (ATA) has been demonstrated with broad-spectrum antimicrobial and antiviral activities. In this study, we implemented ATA treatment in HBV-infected HBV-transfected hepatocytes uncover whether inhibits HBV replication its underlying mechanism. Results DNA levels were significantly reduced, while viral proteins or transcripts not altered. contrast, did further deteriorate DNAs, proteins, transfected by RNase H dead mutant. Moreover, showed inhibitory effect on synthesis lamivudine-resistant mutants. Conclusions potent inhibitor of disrupting the activity polymerase.

rpb۵-mediating protein suppresses hepatitis b virus (hbv) transcription and replication by counteracting the transcriptional activation of hepatitis b virus x protein in hbv replication mouse model

conclusions our study demonstrated that a low dose of rmp could inhibit hbv transcription and replication, which is dependent on the appearance of hbx in vivo. results rpb5-mediating protein could inhibit hbv transcription and replication in groups transfected with the 1.2 wt and hbx. the inhibitory effect disappeared in the 1.2x (-) groups, yet it reappeared in the groups co-transfected with 1...

Inhibition of hepatitis B virus replication by APOBEC3G.

The cytidine deaminase APOBEC3G (A3G) was recently identified as a natural resistance gene that restricts efficient propagation of human immunodeficiency virus and other retroviruses. The enzyme induces massive cytidine to uridine (C3U) deamination of single-stranded retroviral DNA, resulting in DNA degradation or lethal guanine to adenine (G3A) hypermutation (1). Hepadnaviruses, including hepa...

Inhibition of hepatitis B virus replication by interferon requires proteasome activity.

Hepatitis B virus (HBV) replication is inhibited in a noncytopathic manner by alpha/beta interferon (IFN-alpha/beta) and IFN-gamma. We demonstrate here that inhibitors of cellular proteasome activity can block this antiviral effect. These results suggest that a critical component of the IFN-induced antiviral response may be the proteasome-dependent degradation of viral or cellular proteins that...

Anti-Hepatitis B Virus Activity of Flavonoids from Marchantia convoluta

Flavonoids from Marchantia convoluta (MCF) were studied for their ability to inhibit the proliferation of 2.2.15 cells. All concentration of MCF inhibited the secretion of HBsAg and HBeAg in the cultured medium by 2.2.15 cells. Analysis of morphological changes of MCF-treated cells by phase-contrast microscopy revealed a possible mode of action for MCF: inhibition of proliferation of 2.2.15 cel...

RPB5-Mediating Protein Suppresses Hepatitis B Virus (HBV) Transcription and Replication by Counteracting the Transcriptional Activation of Hepatitis B virus X Protein in HBV Replication Mouse Model

BACKGROUND RPB5-Mediating protein (RMP) is associated with the RNA polymerase II subunit RPB5. This protein functionally counteracts the transcriptional activation of Hepatitis B Virus X protein (HBx) by competitively binding to the RPB5; however, the effects of RMP on Hepatitis B virus (HBV) transcription and replication remain unknown. OBJECTIVES The purpose of this study was to investigate...
